Overview

12-[[3-chloro-5-(trifluoromethyl)phenyl]carbamoylamino]dodecanoic acid, also known as 7b, is a synthetic aryl-urea fatty acid derivative developed as a mitochondrial-targeting anticancer agent. It belongs to a series of compounds designed to exploit the metabolic vulnerabilities of cancer cells by targeting the mitochondria. The molecule consists of a 3-chloro-5-(trifluoromethyl)phenyl urea head group attached to a 12-carbon (dodecanoic acid) chain. Its mechanism of action involves the disruption of the mitochondrial membrane potential and the subsequent induction of reactive oxygen species (ROS) production, which triggers apoptosis in cancer cells. Preclinical studies have demonstrated its efficacy against triple-negative breast cancer cell lines, such as the MDA-MB-231 line, where the carbon chain length (C12 in this case) plays a pivotal role in its potency and drug-like properties. While the C13 analogue (7c) in the same series was identified as the most potent lead, the C12 analogue (7b) shows significant activity in research models.
